Antiviral Therapy and the Role of Ccc Dna in Persistent Hbv Infection
نویسنده
چکیده
The main aim of treatment for chronic hepatitis B is to suppress hepatitis B virus (HBV) replication before there is irreversible liver damage. Currently, interferonα (IFN-α) is the only approved treatment for chronic hepatitis B. However, it has limited efficacy, is expensive and is associated with many unpleasant side-effects. In the last 5 years, several new antiviral agents have shown promise in the treatment of chronic hepatitis B, but long-term treatment is needed for sustained antiviral effects. Unfortunately, drug-resistant mutants have been found in some patients who have been on treatment for long durations (> 6 months). The incidence, clinical outcome and biological significance of these mutants will be reviewed.

Point Prevalence of Hepatitis B Virus Infection among Adolescents in Visakhapatnam, India
Introduction: Occurrence of detectable amounts of viral antigen or viral particles in the blood of infected patients with Hepatitis B virus (HBV) is a significant characteristic of HBV infection. Detection of HBV antigen or its DNA among individuals of a community is a crucial factor to know the burden of HBV infection. Hepatitis B surface antigen (HBsAg) is a suitable marker of HBV infection b...
متن کاملComparison of anti-hepatitis B virus activities of lamivudine and clevudine by a quantitative assay.
In this study, we used a quantitative assay to measure the concentration-dependent effects of antivirals on extracellular hepatitis B virus (HBV) DNA as well as on different cytoplasmic and nuclear forms of HBV DNA that participate in HBV replication. HBV recombinant baculovirus, which efficiently delivers the HBV genome to HepG2 cells, was used for this study because (i) antivirals can be admi...
متن کاملTherapeutic strategies for a functional cure of chronic hepatitis B virus infection
Treatment of chronic hepatitis B virus (HBV) infection with the viral DNA polymerase inhibitors or pegylated alpha-interferon has led to a significant retardation in HBV-related disease progression and reduction in mortality related to chronic hepatitis B associated liver decompensation and hepatocellular carcinoma. However, chronic HBV infection remains not cured. The reasons for the failure t...
متن کاملHepatitis B virus (HBV)-specific short hairpin RNA is capable of reducing the formation of HBV covalently closed circular (CCC) DNA but has no effect on established CCC DNA in vitro.
Hepatitis B virus (HBV) covalently closed circular (CCC) DNA is the source of HBV transcripts and persistence in chronically infected patients. The novel aspect of this study was to determine the effect of RNA interference (RNAi) on HBV CCC DNA when administered prior to establishment of HBV replication or during chronic HBV infection. HBV replication was initiated in HepG2 cells by transductio...
متن کاملPresence of intrahepatic (total and ccc) HBV DNA is not predictive of HBV recurrence after liver transplantation.
Previous studies reported that hepatitis B virus (HBV) deoxyribonucleic acid (DNA) can be detected in livers of patients who received transplants for hepatitis B despite the absence of serological markers of HBV recurrence. Quantification of HBV DNA was not performed and presence of covalently closed circular (ccc) DNA was not analyzed in most studies. We aimed to quantify total and ccc HBV DNA...
متن کاملبررسی رابطه بین HBV-DNA و HBeAg و آنزیمهای کبدی در نزد بیماران مبتلا به هپاتیت B مزمن
Background and purpose: Hepatitis B virus (HBV) is a major global health problem. The prevalence of HBV infection varies throughout regions of the world. More than 350 million people live with chronic HBV infection and many different clinical symptoms are associated with it. Long-term complications of HBV infection lead to cirrhosis of the liver and hepatocellular carcinoma. Consequently, 0.5-1...
